Supplements That May Reduce Wegovy Injection Site Reactions: What the Evidence Shows

At a glance

  • Injection site reactions / 1.4% with Wegovy 2.4 mg vs 1% with placebo in pooled adult trials [1]
  • Suspected hypersensitivity / The label says to discontinue Wegovy [1]
  • Bromelain / Intact enzyme reached plasma in 19 men taking 3 g a day, half-life 6 to 9 hours [2]
  • Omega-3 fats / Lower CRP, IL-6 and TNF-alpha across 68 trials; weaker at BMI above 30 in chronic non-autoimmune disease [3]
  • Vitamin D / Skin-applied metabolites reduced allergic skin swelling in mice [5]

What the Wegovy label reports

Wegovy 2.4 mg once weekly was evaluated in three randomized, double-blind, placebo-controlled trials including 2,116 adults with obesity or overweight treated up to 68 weeks, versus 1,261 on placebo [1]. Injection site reactions were reported by 1.4% on Wegovy and 1% on placebo [1]. The label warns that anaphylactic reactions and angioedema have been reported after marketing, and says to discontinue Wegovy if a hypersensitivity reaction is suspected [1]. A prior serious hypersensitivity reaction to semaglutide or any excipient is a contraindication [1].

What supplement research measured

Bromelain is a mixture of protein-digesting enzymes from pineapple [4]. A review describes laboratory and animal evidence of antiedematous and anti-inflammatory activity [4]. In 19 healthy men taking 3 g a day, intact bromelain was detected in plasma, with an estimated half-life of 6 to 9 hours [2]. That shows the enzyme can be absorbed, not that it affects injection site reactions.

A meta-analysis of 68 randomized controlled trials with 4,601 participants found marine n-3 supplementation lowered C-reactive protein, interleukin-6 and tumor necrosis factor alpha [3]. Among participants with chronic non-autoimmune disease, the effect on these blood markers was weaker when BMI exceeded 30 [3]. Blood inflammation markers are not injection site reactions.

In laboratory work, vitamin D3 metabolites suppressed IgE-induced mediator production by mast cells in vitro, and metabolites applied to the skin reduced allergic skin swelling in mice [5]. That work did not test an oral supplement or Wegovy injection site reactions.

What the label says about giving the injection

The label directs subcutaneous injection in the abdomen, thigh or upper arm, once weekly, at any time of day; the time of day and the injection site can be changed without a dose adjustment [1].

Frequently asked questions

How common are injection site reactions with Wegovy?
In the pooled adult weight-management trials, injection site reactions were reported by 1.4% of people taking Wegovy 2.4 mg once weekly and 1% of people on placebo [1].
What did omega-3 or bromelain studies measure?
Omega-3 supplementation lowered CRP, IL-6 and TNF-alpha across 68 trials, with a weaker effect at BMI above 30 among people with chronic non-autoimmune disease [3]. Oral bromelain reached measurable plasma levels in 19 healthy men taking 3 g a day [2].
When is a reaction after a Wegovy injection serious?
The Wegovy label warns that anaphylactic reactions and angioedema have been reported after marketing, and says to discontinue Wegovy if a hypersensitivity reaction is suspected [1]. A prior serious hypersensitivity reaction to semaglutide or an excipient in Wegovy is a contraindication [1].
